    Abstract

Predictive motor control enables animals to compensate for sensorimotor delays and perform timely movements in dynamic environments. Although event timing prediction has been widely investigated in visually and auditorily evoked behaviours, its presence in the vestibular‐motor system remains unclear. Moreover the extent to which predictive learning generalizes across sensory modalities is poorly understood. Recent studies in goldfish have demonstrated that the optokinetic response (OKR), a visually evoked reflexive eye movement, can transition into a predictive mode (pOKR) following exposure to periodic visual motion stimuli. In natural conditions the OKR operates alongside the vestibulo‐ocular reflex (VOR) during head motion to achieve stable vision. Although the VOR rapidly counteracts head movement incompletely, the OKR reduces residual retinal image slip. Notably both reflexive eye movements share neural substrates in the cerebellum and brainstem, which have been demonstrated essential for pOKR acquisition. Here we hypothesized that the VOR could also acquire predictive control, and that predictive ability acquired through visual training as pOKR might transfer to the VOR. Using goldfish we found that periodic vestibular stimulation alone does not induce predictive VOR. However following pOKR acquisition through visual training the VOR evoked solely by vestibular stimulation exhibited predictive behaviour. These findings provide the first evidence that visually acquired predictive control can cross‐modally transfer to the vestibular‐motor system, which itself does not develop prediction through vestibular stimulation alone, highlighting the central role of cerebellar and brainstem circuits in integrating multimodal sensory inputs for adaptive predictive oculomotor behaviour. image

    Key points

Predictive motor control compensates for sensorimotor delays by anticipating periodic events, yet its mechanisms have been studied mostly in visual and auditory systems, leaving vestibular‐motor or cross‐modal prediction largely unexplored.

The vestibulo‐ocular reflex (VOR) and optokinetic response (OKR), which stabilize vision during head motion in the light, share common brainstem and cerebellar circuits.

Predictive OKR (pOKR) emerges after repeated exposure to periodic visual motion, enabling eye movements to decelerate prior to stimulus direction changes.

Our results show that although periodic vestibular stimulation alone fails to induce predictive VOR, visual training that establishes pOKR enables predictive features to emerge in VOR—demonstrating for the first time, cross‐modal transfer of predictive motor control.

These findings reveal a novel form of cross‐modal predictive motor learning and underscore the cerebellar–brainstem circuit's role in integrating multisensory experiences for adaptive motor function.

Prediction of drowsiness based on an objective measure is demanded in machine and vehicle operations in which human errors may cause fatal accidents. Currently we focused on the pupil of the eye which is controlled by the autonomic nervous system, and easily observable non-invasively from the outside of the body. We employed uneventful driving simulation to induce drowsiness of human subjects, and an anti-saccade task to evaluate their cognitive and motor performance. First we confirmed that pupil diameter fluctuates with large amplitude at low frequencies when the subject is aware of his/her drowsiness as reported previously. During this period, the latency of anti-saccade initiation was elongated and varied. We then found that prior to this fluctuation, pupil diameter decreases gradually in most subjects, and they were not aware of sleepiness during this period. We conclude that this monotonic gradual miosis can be a reliable premonitor of drowsiness.

The vestibuloocular reflex (VOR) stabilizes visual image on the retina during head movements. The VOR is under adaptive control in which cerebellum is intimately involved. The VOR research in neuroscience can be divided into 4 different mutually related categories: 1) identification of the neuronal circuitry, 2) identification of the neuronal locus responsible for the motor learning, 3) mechanisms of synaptic and nonsynaptic plasticities at the learning and memory site(s), 4) computational algorithms. This article reviews these aspects in the VOR research, clarifying different mechanisms for the short and long term, and high and low gain VOR motor learning.

Even under the same visual environment, visual image projecting onto the retina and spatially sampled image by retinal photoreceptor array are unique to each individual due to their unique eye optics, spatial arrangement of photoreceptors, and eye and pupil movements. Here we developed a visualization system of spatially sampled image by retinal photoreceptor array of individual subjects, aiming especially at applying to car driver's safety technology. In this study, we evaluated roles of microsaccades that were recently confirmed to be generated during real car driving and known to reflect spatial attention of human and non-human primates.

The optokinetic response (OKR) of the eye is induced to follow a large field visual motion. The gain of the OKR (eye velocity / visual motion velocity) is about 0.6 at 1/8Hz in naive goldfish. If the same periodic visual stimulus is given continuously, the OKR gain increases up to 1.0. In addition to gain increase, goldfish acquires predictive OKR that is named period tuning (PT). Namely, after continuous exposure to a periodic visual motion, goldfish OKR velocity decreases as if it predicts the timing of the change in visual motion direction. Even if the animal is exposed to complete darkness after the acquisition of PT, its eye velocity follows the similar trajectory to that during training for a while without any visual stimulation. This eye movement in the dark may share a common mechanism with optokinetic after nystagmus (OKAN) that is also evoked in the dark after prolonged constant velocity visual stimulation, and considered to reflect characteristics of the velocity storage mechanism (VSM). To investigate the relationship between PT and VSM, we conducted a series of behavioural experiments in human, goldfish, carp, medaka, and zebrafish. We demonstrate that ability to acquire PT is highly dependent on OKAN. This result suggests that VSM is a key mechanism for PT.

In recent years, human errors due to inattentive and sleepy driving are the major cause of fatal car accidents in Japan. To prevent tragic car accidents, it is therefore highly demanded to monitor driver's attention state in a practical way. Recent neurophysiological studies demonstrated that microsaccades (MSCs) reflect spatial attentional states in human and non-human primates. MSC is a fixational eye movement that occurs involuntarily while subjects are fixating on a small visual target. So far, most of researches on MSC have been conducted in unnatural laboratory environments, using controlled artificial visual stimuli under a head fixation condition. Here, we measured eye movements of human subjects during real car driving, and evaluated the relationship between characteristics of MSC generation and road conditions.

In the present study, we evaluated two kinds of characteristics of PT in goldfish: 1) stimulus direction selectivity, and 2) stimulus stop timing dependency. We show that the PT occurs only in the visual motion direction to which visual stimulus was given. Furthermore, the different PT can be induced for the different stimulus directions. These results indicate that the PT is stimulus direction selective. Also, we found that goldfish OKR learns the duration of the visual motion, namely it predicts the end of the visual motion.

In contrast to the horizontal and vertical optokinetic response (OKR) whose behavioral, anatomical and physiological characteristics have been extensively studied, little is known about the torsional OKR. The OKR in response to a step of visual velocity stimulus (step OKR) is a common behavioral test to evaluate dynamic components of the OKR. Horizontal and vertical step OKR consist of direct and indirect components that originate different neuronal pathways subserving the OKR. The indirect component sustains after termination of the visual stimulus and produces continuous eye movements in the complete darkness that is called optokinetic after nystagmus (OKAN). The OKAN of the torsional step OKR has not been well characterized due to the difficulties in measuring roll eye movements generated while the pupil diameter is subject to a significant change. Here we conducted torsional step OKR experiment in human subjects and measured their OKAN by employing the image-processing algorithm that is robust to changes in pupil diameter in measuring roll eye movements. We demonstrate that OKAN after torsional step OKR is clearly generated, but in the opposite direction to the step OKR. This relationship between the directions of OKR and OKAN is contradictory to that of horizontal and vertical counterparts. We also found that subjects were perceiving vection while they were generating torsional OKAN in the dark.

Our recent findings on possible predictors of car driver's sleepiness are reviewed. These predictors employ either the fluctuation of the pupil diameter or a reflexive eye movement called the vestibuloocular reflex or the VOR. In addition, a novel method to detect car driver's inattentive situation utilizing another kind of eye movement called vergence is reviewed. Further, simple and inexpensive implementation of the sleepiness prediction on a smartphone is introduced.

Falling asleep while driving is directly linked with a fatal accident. Therefore, it is important to detect or preferably to predict driver's sleepiness. We have demonstrated that the pupil, and the vestibule-ocular reflex (VOR) can be predictors of one's sleepiness. Here we developed a real-time sleepiness prediction system that initiates a bio-feedback stimulus to a car driver to prevent him/her from falling asleep when sleepiness is predicted. We employed VOR as a predictor, and micro-vibration on the steering wheel as a bio-feedback stimulus. We evaluated the system in driving simulation environment, and demonstrate that the bio-feedback stimulus initiated when sleepiness is predicted can maintain driver's arousal level longer than without the bio-feedback.

Saccades are initiated by phasic pulse-like neuronal activities of brainstem burst neurons. The phasic activities alone cannot maintain the eye position updated after each saccade, resulting in drifting back to the pre-saccadic eye position. Therefore sustained neuronal activities have to be added to the phasic activities to hold the eye position after each saccade. The oculomotor integrator (NI) has been postulated to produce the sustained step-like signal by integrating the pulse-like phasic signal. In goldfish, neurons carrying the signal that is seemingly the output of NI have been identified in Area I. However, it is not yet known how many NIs exist to control two eyes each of which consists of at least two hemi-fields, namely nasal and temporal. Here we employed a novel visual feedback stimulus to test monocularity and eye position dependency of NI. We demonstrate that there are at least 2 NIs each of which is responsible for maintaining temporal eye positions in one eye and nasal positions in the other eye.

The vestibuloocular reflex (VOR) is an adaptive motor control system that maintains compensatory eye movements when growth, aging, injury, etc, may change oculomotor plant dynamics. The cerebellar flocculus is intimately involved in the adaptive VOR control. The VOR has been a popular model system to investigate cerebellar adaptive motor control. Long-term depression (LTD) and long-term potentiation (LTP) at the parallel fiber - Purkinje cell synapses have been thought to be a major underlying mechanism of the cerebellar motor learning. To explore the information processing in the cerebellar neuronal circuitry for VOR motor learning, we constructed an anatomically plausible large-scale neuronal network model of the cerebellar flocculus. The model is composed of spiking neurons with the cerebellar LTD and LTP implemented. In simulation, we confirmed that the model reproduced gain-up and -down VOR motor learning and its frequency selectivity. Analyses of the. properties of the model before and after the VOR training revealed that the cerebellar circuitry formed a band-pass filter in response to vestibular andJor efference copy of eye movement signals after gain-up and -down training. The results suggest that the cause of the frequency selectivity in VOR motor learning is multiple frequency bands implicitly composed in the cerebellar cortical neuronal circuitry tuned by the cerebellar LTD and LTP.

Our body motion is adjusted properly under 1G condition on the earth. Thus changes in gravitational environment have significant impact on our motor control. Further, it may affect on our ability in motor learning and memory. Although some astronauts informally reported difficulties in their body control under microgravity, no quantitative research on this issue has not been conducted. Here we performed the prism adaptation of a hand-reaching task under different gravitational environments. We compared learning and forgetting curves, and memory retention rates of the prism adaptation performed in upright and supine positions, and under 1G and 2G hyper-gravity conditions. We demonstrate that quicker learning, less forgetting and greater memory retention rates are obtained in a supine position and under 2G in comparison with their counter part.

The cerebellum has been implicated to form an inverse dynamic model of a control object. Purkinje cells (PCs), the sole output cell type of the cerebellum, are thought to encode motor commands for their control object that are conformed by cerebellar synaptic plasticity to minimize errors in motor performance. Thus a question arises: Can the PCs that control a biological motor system, such as the eyes, acquire appropriate motor commands to drive a non-biological motor system, such as a direct current (DC) motor? We utilized a vestibulo-ocular reflex (VOR) motor learning scheme. Single unit recording of vestibulo-cerebellar PC's was conducted in goldfish. The desired trajectory of a DC motor was given to the fish in the form of head rotation. The PC firing rate modulated by the vestibular stimulus was converted into a pulse width modulation signal in real time to drive the DC motor. The produced DC motor trajectory was subtracted from the desired trajectory to calculate a performance error. The error signal was fed-back onto the goldfish retina as whole-field visual image motion. This activates VOR motor learning, and lets us evaluate if the PC under recording can improve the performance of the DC motor. The performance error of the DC motor under the VOR motor learning scheme was reduced adaptively. The result suggests that the cerebellum can acquire an internal model of a non-biological motor system, and a single PC is capable of adaptive motor control.

The vestibulo-ocular reflex (VOR) has been a popular model system for the study of cerebellar motor control and learning. The VOR maintains a stable visual image on the retina by counter-rotating the eyes during head movements. Perfect compensation is represented by a gain equalling 1.0 where gain is defined as eye velocity/head velocity. In normal conditions, VOR gain is close to 1.0, however in the laboratory, VOR gain can be modified by using visual-vestibular mismatch stimuli. The cerebellum is intimately involved in VOR gain modification as evident in the firing modulation of its output neurons, Purkinje cells, which closely corresponds to changes in VOR gain. Currently we recorded from cerebellar Purkinje cells during a novel VOR motor learning paradigm in which VOR gains at two different stimulus frequencies are modified in opposite directions. We show that while behavior can be simultaneously modified at both stimulus frequencies, Purkinje cells change their firing modulation at only one.

The opportunity of manned space activity is expected to increase. However, many problems remain unsolved including those relating to autonomic nervous system control in hyper- and hypo-gravity environments. In this study, we evaluated effects of prolonged hyper-gravity on the autonomic nervous system by using pupillary flush responses. We employed a centrifugal accelerator to impose constant hyper-gravity on human subjects along their body axis (max. 3G). It was shown that both sympathetic and parasympathetic nervous systems in a naive subject are activated under constant hyper-gravity, whereas they did not change significantly in a subject who has been exposed to the hyper-gravitational environment many times.

Third, the focusing domain in VR space is shared with collaborators. Finaly, this system is performed in cross-platforms. As a good candidate of applied data, the kinetic evolution of both individual and interconnected status in the cerebellar neural network simulation is chosen. The visualization for pathway of all nodes including intersections between motor learning processes is performed. Our CompleXvision is applicable to recognize holistic and individual behaviors and to identify distinguished behavior nodes without arbitrariness.

In this paper, we have con figured a cerebellar neuronal network model to be used for real time adaptive motor control. The model receives desired trajectories as input, and generates motor commands as output to move the controlled object. The motor output (actual trajectory) is fed-back to the model to calculate the motor error (actual - desired trajectory) which is used for modifying the synaptic weights in the cerebellar neuronal circuitry. We have employed this model as a controller for a DC motor, and evaluated its performance in real time trajectory control of the DC motor. It was demonstrated that the proposed cerebellar controller rapidly improves the performance of the motor when the desired trajectories are a single sinusoid,or band-limited gray noise. It was also demonstrated that the controller can compensate for sudden changes in the weight of motor load and keep accurately tracking the desired trajectory, while a conventional PID controller lost its control in response to the same change in motor load.

Most cerebellar motor learning theories hypothesize that the cerebellar floccular Purkinje cell complex spikes carry retinal slip information which is considered as an error signal to drive the vestibuloocular reflex (VOR) motor learning. It has been suggested the cerebellar Purkinje cell complex spikes also contribute to real time motor control as they modulate during VOR in total darkness. To test whether the floccular Purkinje cell complex spikes can be used as an error signal for VOR motor learning, and whether it carries information as to motor output (eye movements), we continuously recorded complex spike activities in flocculus during acute horizontal VOR motor learning in squirrel monkeys. We show that the complex spikes encode the amount of retinal slip in a non-linear manner in addition to the direction of the retinal slip. Further, we demonstrate that the complex spike firing probability possesses significant amount of information on eye movement right after the occurrence of the spikes.

Spontaneous pupillary oscillation under constant illumination is called pupil nose. In recent years, pupil noise is studied as a subjective measure for assessing exhaustion and sleepiness. Second order statistics such as power spectrum have been widely used to analyze pupil noise and describe its statistical characteristics. However, since pupillary control system is a non-linear system, the level of sleepiness may be reflected in higher-order statistics, such as bispectrum of pupil noise. We recorded pupil noise of human subjects during driving simulation under a sleepiness controlled condition, and calculated its bicoherence. It was demonstrated that there are significant differences in bicoherence of pupil noise depending upon subject's sleepiness.

The light-pupil reflex system has been the object of study in physiological engineering, as a typical example of involuntary biological systems. As regards the variable that actually controls the pupil, however, there has been little discussion of its role in vision. This study notes that the pupil is closely related to the spatial frequency response of the eye's optical system, and discusses the relation between the quality of the retinal image and the pupil size, as determined by the external image and the characteristics of the eye's optical system. By defining the information capacity of the retinal image as the evaluation measure, the quality of the retinal image is represented quantitatively. The relation between the pupil size and the information capacity of the retinal image is examined for various conditions. It is shown that the human pupil is controlled so that the quality of the retinal image is optimized, in the sense of the information capacity of the retinal image for each luminance level, when the external image is a natural image. On the other hand, when the external image is not a natural image, or when the characteristic of the eye's optical system or the relation between the visual cell arrangement and the pupillary diameter differs from the actual situation, it is not necessarily true that the pupil optimizes the quality of the retinal image. The above results suggest that the function of the pupil in vision is optimization of the quality of the retinal image. It is also suggested that the adjustment function of the pupillary diameter is acquired under the constraints of the external image and the hardware of the eye. © 2003 Wiley Periodicals, Inc.

In this study,we developed a cardiovascular model involving gravitational effects to investigate cardiovascular dynamics during parabolic flight. This model is based on DeBoer's beat-to- beat model whose validity has been verified for subjects under the resting state in normal gravity environment.Taking into account the fluid shift caused by gravitational change,we incorporated two gravitational effects into the model. One is blood pressure changes on baroreceptors at carotid sinus, and the other is venous return to the right heart and redistribution of blood at the left ventricle. Moreover,a method of estimating model parameters from recorded cardiovascular signals was proposed.Simulation results show that when recorded gravity and respiration data are used as input to the model, the model output and the experiment results are roughly the same.In conclusion,it was found that when these gravitational effects are incorporated into the model, the cardiovascluar dynamics during parabolic flight is well explained.

In this study, we simulated vestibuloocular reflex and tilt translation ambiguity using realistic cerebellar model. In the results, we revealed that the plasticity at the synapse between parallel fiber and Purkinje cell tuned motor related input signal to achieve goal of performance in the motor control during learning. Furthermore, our cerebellar model can resolve tilt translation ambiguity and reproduced neuronal activity that shown in experimental study. Those results also indicate the versatility of cerebellar motor learning.

We first discovered various basic characteristics of predictive optokinetic response (OKR) in goldfish, then demonstrated that the cerebellum is required for the acquisition of these behaviors. We also configured a mathematical model of predictive OKR based upon known and newly discovered anatomy and physiological evidence. Furthermore, we implemented a major part of the model onto FPGA to enable real-time execution of the model, and applied it to real-wold motor control as an adaptive predictive controller.

The aim of the research was to apply knowledges on neural mechanisms of biological adaptive control to various engineering problems. To achieve the goal: 1) We conducted neurophysiological experiments in goldfish while they were learning new eye movement control. We recorded single unit activities from the cerebellum and brainstem areas known to be involved in the oculomotor control, and identified their characteristics during the eye movement learning. 2) Based upon the results and accumulating related evidence from other labs, we configured a mathematical model of eye movements in which the cerebellar-brainstem loop is explicitly described, and demonstrated its validity. 3) By simplifying the model so that it runs in real time, we employed the model as an adaptive controller of robots, and demonstrated its validity in actual robot control experiments. 4) We further developed a cerebellum-machine interface that enables adaptive robot control by goldfish cerebellar neuronal activities.

School of Med.)
