    We developed a novel approach for generating rat offspring using rat embryonic stem (ES) cell-derived sperm produced in mice with the blastocyst complementation method. By optimizing culture conditions, we established naïve male rat ES cells from two transgenic rat strains expressing EGFP and Venus fluorescence, respectively. The pluripotency of these cells was confirmed by the formation of germline chimeras. These ES cells were then injected into blastocysts of germ cell-deficient mice, which resulted in chimeric mice with the ability to produce rat-derived sperm. Histological analysis confirmed the presence of seminiferous tubules and spermatozoa, which are morphologically characteristic of rats, in the chimeric testes. To evaluate the fertilization potential of the chimeric mouse sperm, we performed intracytoplasmic sperm injection (ICSI) to rat oocytes and successfully produced viable offspring carrying ES cell-derived traits. This method eliminates concerns regarding host cell contribution, as all sperm in the chimeras originate from rats, enabling the use of nonfluorescent cells. Furthermore, the absence of competition with host cells is expected to enhance sperm production efficiency. By utilizing germ cell-deficient mice as recipients, this approach offers a cost-effective and efficient strategy for generating genetically modified rats, addressing key limitations in rat ES cell-based genetic engineering.

    We investigated potential germline‐specific radiosensitive biomarkers in the testes of large Japanese field mice (Apodemus speciosus) exposed to low‐dose‐rate (LDR) radiation after the Fukushima accident. Fukushima wild mice testes were analysed via RNA‐sequencing to identify genes differentially expressed in the breeding and non‐breeding seasons when compared to controls. Results revealed significant changes during the breeding season, with Lsp1 showing a considerable upregulation, while Ptprk and Tspear exhibited significant reductions. Conversely, in the non‐breeding season, Fmo2 and Fmo2 (highly similar) were significantly upregulated in radiation‐exposed Fukushima mice. qPCR analysis results were consistent with transcriptome sequencing, detecting Lsp1 and Ptprk regulation in the testes of Fukushima mice. While differences in gene expression were observed, these do not imply any causal association between the identified biomarkers and chronic LDR exposure, as other factors such as the environment and developmental age may contribute. This study provides valuable insights into the reproductive biology is affected by environmental radiation and highlights the value of assessing the effects of chronic LDR radiation exposure on testicular health in wild mice.

    Abstract

    This study investigated the effects of chronic low-dose-rate radiation exposure on gene expression related to immunological defense mechanisms and epigenetic regulation in the testis of male mice. The study found that radiation exposure (4.5 mGy/hr until 4000 mGy) led to a significant decrease in the expression of genes involved in epigenetic regulation and a significant increase in the expression of several genes linked to innate and acquired immunity, suggesting a noticeable effect on the immune response. In addition, there was a significant increase in the expression of SRY-box transcription factor 9, which plays a crucial role in testicular development, indicating a response to irradiation. These findings provide insights into the biological effects of chronic low-dose-rate radiation exposure on innate immunity and epigenetic regulation in the testis.

    Since the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, great attention has been paid to the impact of chronic low-dose-rate (LDR) radiation exposure on biological systems. The reproductive system is sensitive to radiation, with implications connected to infertility. We investigated the testis ultrastructure of the wild large Japanese field mouse (Apodemus speciosus) from three areas contaminated after the FDNPP accident, with different levels of LDR radiation (0.29 µSv/h, 5.11 µSv/h, and 11.80 µSv/h). Results showed good preservation of the seminiferous tubules, comparable to the unexposed animals (controls), except for some ultrastructural modifications. Increases in the numerical density of lipid droplet clusters in spermatogenic cells were found at high levels of LDR radiation, indicating an antioxidant activity rising due to radiation recovery. In all groups, wide intercellular spaces were found between spermatogenic cells, and cytoplasmic vacuolization increased at intermediate and high levels and vacuolated mitochondria at the high-level. However, these findings were also related to the physiological dynamics of spermatogenesis. In conclusion, the testes of A. speciosus exposed to LDR radiation associated with the FDNPP accident showed a normal spermatogenesis, with some ultrastructural changes. These outcomes may add information on the reproductive potential of mammals chronically exposed to LDR radiation.

    Wild large Japanese field mice (Apodemus speciosus) responses to cyclic seasonal changes are associated with physiological and behavioral changes. However, the detailed regulation of oogenesis in the ovary during the seasonal reproductive cycle in wild large Japanese field mice has not been studied. We assessed the dynamics and changes in ovarian morphology and hormone concentrations associated with reproductive seasonality throughout the year. The stages of the ovarian morphological breeding cycle of wild large Japanese field mice were classified as breeding, transition, and non-breeding periods during the annual reproductive cycle. Measurement of blood estradiol concentrations throughout the year showed that the levels in September and October were higher than those in other months. It is presumed that follicle development starts from a blood estradiol concentration of 38.4 ± 27.1 pg/mL, which marks a shift from the transitional season to the breeding season, followed by the transition to the non-breeding season at 26.1 ± 11.6 pg/mL. These results suggest that seasonal follicle development in wild rodents is correlated with estradiol regulation. We consider this species to be an alternative animal model for studying seasonal reproductive changes and the effects of environmental changes.

    The intergenerational effects from chronic low-dose exposure are matters of concern. It is thus important to elucidate the radiation-induced effects of germ cell maturation, fertilization and embryonic development. It is well known that DNA methylation levels in CpG sites in gametes are reprogrammed in stages during their maturity. Furthermore, the binding of Izumo on the surface of sperm and Juno on the surface of oocytes is essential for fertilization. Thus, there is a possibility that these genes are useful indicators to evaluate fertility in mice after irradiation exposure. Therefore, in this study, we analyzed global DNA methylation patterns in the testes and gene expression of Izumo1 and Izumo1r (Juno) in the gonads of mice after neonatal acute high-dose ionizing radiation (HDR) and chronic low-dose ionizing radiation (LDR). One-week-old male and female mice were irradiated with a total dose of 4 Gy, with acute HDR at 7 days at a dose rate of 30 Gy/h and LDR continuously at a dose rate of 6 mGy/h from 7 to 35 days. Their gonads were subsequently analyzed. The results of global DNA methylation patterns in the testes showed that methylation level increased with age in the control group, the LDR group maintained its DNA methylation level, and the HDR group showed decreased DNA methylation levels with age. In the control group, the gene expression level of Izumo1 in the testis did not show age-related changes, although there was high expression at 100 days of age. However, in the LDR group, the expression level recovered after the end of irradiation, while it remained low regardless of age in the HDR group. Conversely, gene expression of Izumo1r (Izumo1 receptor) in the ovary decreased with age in the control group. Although the gene expression of Izumo1r decreased with age in the LDR group, it remained low in the HDR group. Our results indicate that LDR can induce different DNA methylation patterns, and both high- and low-dose radiation before sexual maturity might affect gametogenesis and fertility.

    実験動物における生殖組織の評価系を確立するためにマウスに放射線を照射し、被ばく後の生殖細胞形成機構を明らかにするために、幼児期被ばくと青年期被ばくを想定した被ばく時年齢、生殖巣の回復できる線量の検討を行った。具体的に、8日齢(幼児期被ばく)、50日齢(青年期被ばく)のICRマウスに対して1,2, 4 Gy照射後、つがいで飼育し、生殖細胞形成が観察されるであろう時期にサンプリングを行った。しかしながら飼育中に多くの個体が死亡したため、放射線照射や飼育条件について検討を行う必要があった。
    一方、予備検討の結果、急性照射や慢性照射後において、照射終了後にマウスの精巣重量が回復することから、精巣における回復メカニズムを検討することができると考えた。精巣の維持に関わるといわれている遺伝子に着目して、上記実験におけるマウスの精巣の遺伝子発現量を測定した。この遺伝子の発現量は対照群、2Gy照射群において加齢に伴い増加していたが、4Gy照射群では2.4倍に減少した。さらに、青年期被ばく群では、照射線量が高くなるにつれ、遺伝子発現量も増加傾向にあった。この遺伝子は、加齢だけでなく照射後においても遺伝子発現の増幅傾向が見られることから精巣の維持、回復に関わっている可能性が考えられ、文献情報と一致している。
    さらに、精子細胞膜に発現するIzumo1の遺伝子発現量は、放射線の影響評価の指標として有効であるため、放射性物質汚染地域のアカネズミの精巣の影響解析に適用できるか検討した。まずマウスのIzumo1遺伝子からプライマーを設計した。これを用いてアカネズミの精巣cDNAを鋳型とし、PCRにより増幅した。増幅されたDNA断片のクローニングおよび塩基配列を決定したところ、得られた約450bp、約650bpの断片はマウスと高い相同生を示した。

    糖尿病と放射線被ばくは共に炎症を引き起こすが、糖尿病に由来する炎症と、放射線被ばくに由来する炎症の相乗/相加的影響は不明であり、特に、晩発性の影響については知見が乏しい。そこで、本研究では、二つの2型糖尿病モデルマウスに非致死線量のX線を照射し、慢性炎症である「糖尿病」と急性から慢性の炎症を引き起こす「放射線被ばく」の二重ストレスによる糖尿病性病変、白内障、糖代謝機能および消化管障害における相乗的または相加的影響を解析する。さらに、放射線照射後の2型糖尿病マウスの精巣および精子を解析し、放射線被ばくによる顕著な精巣退縮のメカニズムを明らかにする。これらの研究により、放射線被ばくの後発性影響のリスクの再評価を行い、放射線被ばくによる健康被害の軽減に貢献することを目的とする。
    当該年度は新型コロナウイルスのパンデミックによりモデルマウスの購入が困難となったため、実験計画の進行に大きな支障をきたした。そのため、解析に必要となる解析技術の最適化に取り組み、細胞質分裂阻害微小核法の培養時間の最適化、マウス生殖細胞の損傷解析のための技術開発、生殖巣における細胞死や細胞増殖解析法の確認などを行った。
    細胞質分裂阻害微小核法による染色体異常の解析においては、研究成果を国際生物線量評価会議(IABERD)が主催する国際学会EPRBioDose2022 onlineで共同研究者であるGoh氏が発表し、Poster Awardsを受賞した。当該発表演題はヒト末梢血を用いた研究であるが、マウス脾細胞培養での有用性を確認した。

    The objective of this study was that the finally generating cattle and rat germ cell in the mouse by the Cas9 knockout and/or gene targeting method combined with interspecies blastocyst complementation. In this experiment, we found that (1) The Nanos gene family is required for germ cell development in diverse organisms. We successfully generated Nanos3 knockout mouse, results in the complete loss of germ cells in both testis and ovary. (2) GATA4 gene encodes a member of the GATA family of zinc-finger transcription factors. Hetero knockout mouse produced by crossing GATA4flox/flox mouse with GATA4flox/+;Vasa-Cre mouse. This hetero knockout mouse was maintained reproductive function, but showed a marked decreased in the number of litters compared to the wild type. In future, these results could be useful for the generation of not only animal but also human germ cells and organs into chimeric animals produced by xenogeneic Blastocyst complementation.

    We aimed to investigate the effect of exporting radiation for 6 months and 2 years in the ex-evacuation zone, a 20-km radius region surrounding the FNPP, on whole genome exome mutation of offspring associated with radiated bulls. As a result, there were no hereditary phenotypic alterations in the whole genome exome regions of calves associated with paternal exposure to ionizing radiation. Furthermore, we analysed transgenerational effects on calf on the blood plasma metabolome in order to detect alterations associated with paternal exposure to radiation. There were no significant transgenerational effects on the blood plasma metabolome in calf obtained by AI using sperm from LDR-radiated bull in the FNPP ex-evacuation zone. In calves produced by using sperm of Fukushima bulls, the radiation effects were extremely low. Thus, the results of our study would contribute to the reconstruction of the Fukushima livestock industry and facilitate devising food safety and security measures.

    We have constructed an archive system of organs affected by the Fukushima Daiichi Nuclear Power Plant accident and obtained the following results. Two years after the accident, almost only radioactive cesium was detected in all organs examined. Oxidative stress related molecules in cattle plasma were correlated with internal dose-rate. DNA double strand breaks in cattle lymphocytes were higher than non-affected controls. Large Japanese field mice were significantly exposed to both gamma- and beta-rays. The turnover of their spermatogenesis was accelerated. However, no abnormalities was detected in mature sperms including cattle. Bone marrow of wild Japanese macaques tended to be hypoplastic, but no abnormality was found in peripheral blood cells. Although no significant changes have been observed, internal dose rate in monkeys is still high. Therefore, it is necessary to continue detailed study to know biological effects of long-term exposure to low dose-rate radiation.

    The creation of an efficient technique for producing transgenic chicks may lead to industrial applications in agriculture and biopharmacy; moreover, it will advance our understanding of avian biology itself. The objective of this study was to investigate the method for producing transgenic avian species by injected piggyBac transposon constructs into blastoderm after laying the egg. Consequently, when 2.5ng/ul piggyBac transposase plasmid was injected into the egg blastoderm, it could be introduced for gene transferred transgenic chicks.

    ラットは、生体機能モデルや疾患モデルとして利用される重要な実験動物である。しかし、多種多様なラット遺伝資源の効率的確保、安定した供給や保存において極めて重要な技術である精子の凍結保存技術は未だ実用化に至っていない。それ故、ラット精子の凍結保存技術の開発は急務となっており、その技術が実用可能になれば、開発の意義は計り知れない。そこで本研究では、極めて困難であるとされるラット精子凍結保存法の開発と凍結精子による体外受精技術の確立を目的として実験を行った。その結果、1)概知のラクトース卵黄保存液に代わる新たな精子の凍結保存液として基盤となる、ラフィノース・卵黄-修正クレブス-リンガー重炭酸液(modified Krebs-Ringer Bicarbonate;mKRB)保存液を開発した。2)ラット精子のエネルギー利用と供給に着目した、乳酸、およびエネルギー源であるATPを添加したラフィノース・卵黄-mKRB凍結保存液を改良した。3)精子運動活性化作用を有する物質に着目し、精子内のcAMP濃度を上昇させる効果のあるdbcAMPをさらに添加したdbcAMP・ATP・乳酸・ラフィノース・卵黄-mKRB保存液を開発した。4)新鮮精子を用いた体外受精および培養法において、ATPおよびdbcAMPを添加した受精培地を改良・開発し、従来の方法に比較して、高い受精率と胚の発生率を得ることが可能になった。さらに、凍結保存した精子においても、開発した受精培地を用いることにより、受精可能であることを証明した。以上、本研究において開発されたラット精子凍結保存法は、精子の凍結保存に有効な技術であるはかりではなく、保存精子を用いた体外受精においても有効な技術であることを示した。これらのことから、本研究の新たな知見と技術は、重要な実験動物であるラット遺伝資源の効率的維持、保存や安定した供給のため貢献するものである。
