    Phedimus individuals from Mt. Daeam, once referred to as Phedimus sikokianus, exhibit certain morphological characters that are unique within the genus. Phedimus is one of the most notorious groups for taxonomic problems due to the high morphological variation found in leaf shape, stem numbers, phyllotaxis and seed structure. Taxa in Phedimus also easily hybridize, further leading to taxonomic confusion. To carefully confirm the identity of the putative new species from Mt. Daeam, we examined morphological characters from ~100 herbarium sheets of six closely related Phedimus species. A molecular phylogenetic approach was also employed to delimit the species boundary and infer the phylogenetic relationships among the seven Phedimus species, including the species from Mt. Daeam. Both morphological and molecular phylogenetic results indicated that the species found on Mt. Daeam is a new species that is more closely related to P. middendorffianus and P. takesimensis than to the remaining four Phedimus species. Here, we provided a full description of the new species P. daeamensis as well as an updated key for the seven Phedimus species examined.

    A combination of field and herbarium surveys recorded a total of 34 plant species, one variety of aquatic vascular plant, two aquatic moss species and two charophyte species in Lake Ozenuma and the wetlands surrounding the lake. As a result of the surveys, Acorus calamus L., Typha latifolia L., Potamogeton octandrus Poir. and Schoenoplectus nipponicus (Makino) Soják were newly recorded in the area. However, the former two species are presumed to have occurred in the region in the early of the 2000s. Further, four of the species that were previously recorded as being distributed in the region were considered to have been misidentified; i.e. specimens previously identified as P. nipponicus Makino, S. tabernaemontani (C.C.Gmel.) Palla, Myriophyllum spicatum L. and Utricularia intermedia Heyne were identified as P. gramineus L., S. lacustris (L.) Palla, M. verticillatum L. and U. ochroleuca R.W.Hartm in this study, respectively.

    A small semiartificial pond, revealing high species richness with respect to the aquatic macrophytes, was studied in the Aomori Prefecture, Japan. The pond had been completely formed ca. 10 years previously after excavation into a wet bog. To identify and evaluate its aquatic macrophyte flora during 2017-2018, we listed the species of aquatic vascular plants and charophytes in the pond and compared this list with those from the natural Japanese lakes using a database of aquatic flora in Japan. Two species were identified by molecular analyses because they lacked any reproductive organs, which were necessary for identifying the species level. We found a total of 57 taxa of aquatic macrophytes including 15 Red List species in Japan or Aomori Prefecture. Comparing the flora list with those from the 66 natural Japanese lakes surveyed since 2001, the pond was ranked fourth in the order of diversity (based on species richness) and rarity (based on numbers of Red List species). Therefore, the pond is an important aquatic environment in terms of aquatic macrophyte conservation in Japan. Interestingly, for a pond with such high species diversity, it is relatively small (0.14 km(2)), semiartificial, and relatively recently formed.     A new locality for Potamogeton lucens L. (Potamogetonaceae) was recorded from a pond in Aomori Pref., Tohoku District, Japan. In Japan, this species is critically endangered, and the sole remaining naturally established population was previously known only from one other locality. In June 2017, we found fragments of a submerged plant species that resembled P. lucens. We conducted fi eld surveys and identifi ed it as P. lucens based on morphological measurements, nuclear and chloroplast molecular analyses, a herbarium specimen survey, and pollen fertility. Although we could not reveal its population size and distribution in the pond, at least an established population exists because many fragments and a plant with developed roots were collected. This new locality of P. lucens represents the new northern limit of the species in Japan, which is over 500 km away from its nearest distributional record. As developments are now in progress around the pond area, urgent protection by a conservation program is needed.

    We investigated aquatic flora of the Kumeda-ike pond in Kishiwada City, Osaka Prefecture in September, 2017. With the help of this study, 12 species of aquatic vascular plants and one stonewort species were recorded including three endangered species. The most dominant species were Myriophyllum spicatum L., Stuckenia pectinata (L.) Börner, and Potamogeton pusillus L.. Electric conductivity of the Kumeda-ike pond was 2.57 mS/cm. We attempted to reconstruct the past aquatic flora of the Kumeda-ike pond using records of specimens, literature, and personal observation. It was revealed that various aquatic plants were present in the 1950s according to the specimen records. These aquatic plants disappeared perhaps by eutrophication of pond in the 1980s according to the literature record, and an invasive species, Elodea nuttallii (Planch.) St. John, dominated the pond in the early 2000s according to personal observation records. Thereafter, the dominant species has been replaced by M. spicatum L. since around 2007. The aquatic flora of the Kumeda-ike pond has possibly changed with varying water quality.

    Mr. Shigetaka Hamashima donated his aquatic plant specimens to Osaka Museum of Natural History (OSA) on December 2010. This collection consists of 28 families, 129 taxa, and 1028 specimens collected from 1967 to 2010. These specimens were collected at 26 prefectures, mainly in Tokai region : Aichi Prefecture (542 specimens, 52.7%), Gifu Prefecture (203 specimens, 19.7%), and Mie Prefecture (124 specimens, 12.1%). These specimens are important collections for knowledge of wetland natural history in Tokai region. In this paper, we list the aquatic specimens donated to OSA and the related literature of Mr. Hamashima.

    We identified Sagittaria plants distributed in Lake Yonuma (Iwate Prefecture, Japan) via DNA sequencing and also studied the morphology, distribution, and habitat at Lake Yonuma during 2012 and 2015. In addition, we cultivated plants in an outdoor aquarium. The internal transcribed spacer (ITS) region sequence of these unidentified Sagittaria plants completely matched that of S. natans Pall. from Hokkaido, Heilongjiang, Inner Mongolia, Sakhalin, and other GenBank data. In the field, almost all plants sprouted from vegetative propagules and tubers remaining on the parent plants. Some floating-leaved plants had stolons and new tubers. Although no plants flowered, under natural or cultivated conditions, genetic and morphological characteristics indicate that the plants were S. natans. The population in Lake Yonuma is a remnant population of S. natans in Japan and is located at the southeastern extent of the species distribution worldwide. This population of rare plants requires urgent protection.

    Nuphar submersa (Nymphaeaceae) is a critically endangered freshwater macrophyte indigenous to central Japan, with only four small extant populations represented across its entire range. We investigated the genotypic and genetic diversity as well as the genetic structure of all extant individuals of N. submersa based on analysis of 15 microsatellite loci. Among 278 individual ramets, 52 multilocus genotypes were detected: 30 genotypes in Nikko City (NIK), 18 in Nasukarasuyama City (NAS), 3 in Mooka City (MOK), and 1 in Sakura City (SAK). The average number of alleles per locus ranged from 1.20 to 1.93, whereas the observed and expected heterozygosities ranged from 0.11 to 0.33 and from 0.10 to 0.24, respectively. With the exception of SAK, all populations contained multiple clones, but our results indicated low levels of within-population genetic diversity. The populations NIK and NAS comprised few large or middle-sized genets and many small genets. The populations NIK and NAS were suggested to comprise large old, old fragmented, and/or young small genets resulting from seedling establishment. All four populations were differentiated, and gene flow between the populations was restricted (average level of gene flow (Nm) = 0.122, G' (ST) = 0.639). Of the total genetic diversity, 67.20 and 9.13% were attributable to inter- and intra-population diversity, respectively. STRUCTURE analysis revealed two or three well-differentiated groups of populations. Cluster I comprised one population (NIK) and cluster II comprised the remaining populations at K = 2. The populations NIK, NAS, and the remaining populations were assigned to clusters I, II, and III, respectively, at K = 3. For conservation practices, we recommend that each cluster be regarded as a different management unit. We further suggest that artificial gene flow among MOK and SAK populations is an appropriate option, whereas NIK should not be reinforced with genotypes from the remaining populations.

    We evaluated the actual distribution, present status, and number of remnant individuals of Korean Sagittaria aginashi (Alismataceae) based on herbarium specimens and field surveys. We also reidentified Korean S. aginashi by analyzing sequences of the internal transcribed spacer (ITS) region. We found nine specimens of S. aginashi in herbaria from four localities housed at the Osaka Museum of Natural History (OSA) and at the Korea National Herbarium (KH). During our field research, we could not confirm the current statuses of two collection localities (Taereung and Ansan-si), though this was not the case for Ulsan-si. In addition, we found two new localities in Ulsan-si and Yangsan-si. These three remnant populations are distributed in wetlands in mountainous areas (elev. 480–740 m). With regard to the number of flowering and immature individuals in the three localities, there were <50 and <2,000, respectively, in total. ITS sequences demonstrated that the sequences of Korean and Japanese S. aginashi are uniform and distinct from other Sagittaria species. These results indicate that S. aginashi is a rare and threatened species in Korea. It should be listed as an endangered species on the Korean Red List and requires urgent protection by conservation programs, including the extensive surveys of other possible natural habitats.

    Premise of the study: Nuphar species (Nymphaeaceae) are representative aquatic plants in irrigation ponds in Japanese agricultural ecosystems. We developed 15 polymorphic microsatellite markers for N. japonica and confirmed their utility for its close relatives N. oguraensis var. akiensis and N. xsaijoensis, which originated from natural hybridization between N. japonica and N. oguraensis.Methods and Results: Genetic variation was characterized in 15 polymorphic loci in three populations of N. japonica. The average number of alleles per locus was 3.47 (range = 2-9; n = 32), and the average expected heterozygosity per locus was 0.84 (range = 0.5-1.0); 11 loci were amplified in N. oguraensis var. akiensis and 15 in N. xsaijoensis.Conclusions: The polymorphic microsatellite markers developed in this study will be useful for investigating the levels of genetic diversity within remnant populations of Nuphar taxa and could provide a valuable tool for conservation genetics of these taxa.

    The reintroduction of ex situ conserved individuals is an important approach for conserving threatened plants and reducing extinction risk. In this study, we elucidated the effects on the genetic diversity of wild populations of Vincetoxicum pycnostelma Kitag. [=Cynanchum paniculatum (Bunge) Kitag.] by modelling the genetic consequences of reintroducing plants using the germinated seeds of herbarium specimens. This semi-natural grassland herb is threatened in Japan. First, we tested the germinability of seeds from herbarium specimens collected from nine sites in Kinki and Tokai districts, Japan (one specimen per site, total 206 seeds). Next, we analysed the genetic diversity and structure of germinated seedlings and the current wild individuals using nine polymorphic microsatellite markers. Germination was observed for 38 seeds (18.4 %) from four specimens collected 3-18 years prior to the study. Although the genetic diversity of the specimens' seeds was lower than that of the wild population because of the small sample size, the seedlings from specimens taken from three sites had unique alleles that did not exist in the wild populations. Consequently, viable herbarium specimens' seeds with unique alleles could be useful resources for recovering the genetic diversity in threatened wild plant populations.

    Here we report a previously unrecorded species of Korean Myriophyllum L. (Haloragaceae). This taxon, M. oguraense Miki has been regarded as a Japanese endemic for some 60 years. Myriophyllum oguraense and its closely related M. verticillatum L. share the characteristic of having pectinate emergent leaves that are similar in shape to, but much smaller than, the submerged leaves. However, the primary characteristic that differentiates these taxa is the turion shape (club shaped in M. verticillatum and linear in M. oguraense). The common name, ‘Gin-dong-a-mul-su-sae-mi’ was also newly given considering its characteristic turion shape. Photographs and a key to Korean Myriophyllum species are provided in addition to complete descriptions including information on nomenclatural types, distributions and specimens examined.

    Nuphar submersa and N. ×fluminalis (Nymphaeaceae) are critically endangered macrophytes found in central Japan, comprising only four populations each. In recent years, gathering for horticultural trade has damaged the remnant populations. We investigated the species and localities involved in trading plants of these species and Nuphar plants named "Nagaba-benikohone" based on morphological research and analysis of 15 microsatellite loci. We bought 10 plants from two garden stores. Our morphological research identified a plant named "N. submersa" sold by Store T (T1) as N. submersa and nine plants sold by Store C (C1-C9) as N. ×fluminalis. In these 10 plants, two multi-locus genotypes were detected. Referring to the genotype data for N. submersa and N. ×fluminalis, the genotype of T1 corresponded to NIK-25 (N. submersa), and the other nine plants (C1-C9) corresponded to SAN-10 (N. ×fluminalis). The low genotype probability (GP) indicated that the traded plants and wild plants were the same genets and gathering had occurred in populations NIK and SAN. Since our preliminary genetic identification of trading in Nuphar was published, there have been no trades on the web sites. Genotype data based on all remnant individuals will be useful for assisting in preventing the gathering and trade of these plants.

    We developed and characterized 13 microsatellite markers for Nuphar submersa, a critically endangered freshwater macrophyte of central Japan. In all the studied populations, ten out of isolated 13 loci showed polymorphism. The 13 loci showed one to five alleles per locus, with observed and expected heterozygosities ranging from 0 to 0.40 and from 0 to 0.67, respectively. Between 5 and 11 of the 13 loci were successfully amplified across the six other Nuphar taxa. The markers described here will be useful for a conservation genetic study of the species and informing the development of effective conservation programs of N. submersa and related species. © 2011 Springer Science+Business Media B.V.

    The genetic relationships of Nuphar japonica, N. oguraensis, N. subintegerrima and two morphologically intermediate plant groups (J-O and J-S) found in central to western Japan were investigated. Allozyme analyses (analysis of allele frequencies and principal coordinate analysis, based on shared allele distance among the 162 multi-locus genotypes) showed the three Nuphar species to be distinguished, and indicated that the intermediate plant groups were genetically intermediate either between two or among three of these species. Natural hybridization and introgression were found to have occurred among these three species in central to western Japan, producing genetically varied intermediate plant groups. Many of the intermediate plants (74.4% of the total numbers of genotypes of the intermediate plants) did not show additive combinations of parental allozymes, which were expected in F-1 hybrids. The absence of characteristic parental bands suggested that sexual reproduction had continued among the hybrids beyond the F-1. (c) 2007 Published by Elsevier B.V.

    We investigated morphological and allozyme variation and pollen viability in 22 populations of Nuphar japonica, N. pumila, and unidentified intermediate plants in Hokkaido, Japan, to assess hybridization between the two species and the potential existence of homoploid hybrid speciation. A phenogram based on cluster analysis using 15 morphological characters revealed three cluster groups. Groups 1 and 3 had distinctive morphological characteristics and corresponded to the published descriptions of N. japonica and N. pumila, respectively, whereas Group 2 showed intermediate values in most characteristics. In our allozyme study, many morphologically intermediate plants showed additive combinations of species-specific alleles from the two species. Pollen viability was significantly lower in percentage in intermediate populations than in the two species. Intermediate populations were found within the area of overlap between the ranges of the two species. On this evidence. we concluded that intermediate plants represent hybrids between the two species. Four hybrid populations showed different relationships between morphology and pollen viability. Hybrids in artificially disturbed populations had discontinuously intermediate morphology between their parental species and were completely fertile in some cases. It is probable that ecological selection has contributed to the discontinuous morphological patterns. As a result, homoploid hybrid speciation may be occurring in these Nuphar hybrids.     A new species of Nuphar (Nymphaeaceae), N. submersa, from Tochigi Prefecture, central Japan, is described. Nuphar submersa is characterized by its submerged habit and narrowly oblong-triangular leaves that lack a sinus. It differs from both N. japonica and N. oguraensis in the color of the anthers and fruits. A phylogenetic study based on Amplified Fragment Length Polymorphism (AFLP) fragments showed that both N. submersa and N. oguraensis were sister taxa, but they were well separated phylogenetically.

    Morphological variation in 52 populations of Nuphar japonica DC., N. subintegerrima (Casp.) Makino, N. oguraensis Miki and unidentified intermediate plants were investigated during a revision of Nuphar (Nymphaeaceae) in Japan. The phenogram of cluster analysis based on 15 morphological characters revealed five cluster groups (A〜E). Clusters A, C and E had some remarkable characters and corresponded well to the description of N. japonica, N. oguraensis and N. subintegerrima sensu stricto, respectively. In contrast, Clusters B and D showed intermediate values in most of their characters. It was concluded that the intermediate plants, which usually had been identified as N. subintegerrima, were not conspecific with N. subintegerrima s. s. and that there were two different intermediate groups in central to western Japan. Morphological relationships suggested that the two intermediate groups were of hybrid origin between N. japonica and N. oguraensis and between N. japonica and N. subintegerrima, respectively. Results from the present study proved that populations of N. subintegerrima s. s. are limited in number and distribution, and that the species is one of the most endangered plants in Japan.

    秋の草原はイネ科の植物で彩られます。今月の表紙の植物、オガルカヤは明るい草地に生える多年草です。花序は二つが対になって葉腋につきます。この姿がスズメが飛んでいる姿に似ていることからスズメカルカヤの別名もあります。ちなみにハーブとして使用されるレモングラスもオガルカヤ属です。吹田市にて2011年10月21日撮影。

    博物館の標本庫には100年以上前のものから現在に至るまで、数多くの植物標本が収められており、都市化などにより現在では失われてしまった集団の標本も残されている。このような標本から種子や胞子を採集し、撒きだすことによって失われた集団を復元したり、遺伝的に劣化した集団の遺多様性を回復させたりすることができる可能性がある。そこで、本研究では標本の種子から集団を復元することが可能な種をリストし、その状態を明らかにするために、発芽試験と種子の染色試験を行った。
    調査対象種は絶滅危惧植物を中心に選び、大阪市立自然史博物館(OSA)において81種255点の標本から種子・胞子を得た。発芽試験は段階温度法を用いて行い、未発芽種子はテトラゾリウム1%溶液に浸けて胚の酵素活性の有無を確認した。
    発芽試験の結果、18種33点の標本種子からの発芽が確認できた(1番古いものは1989年採集のミヤコグサ)。また、テトラゾリウム染色試験によって未発芽種子の胚が赤色に呈色し、酵素活性があった標本は47種74点であった(1番古いものは1925年採集のヒメヒゴタイ)。未発芽の生存種子は発芽条件を変えたり、植物ホルモンを添加したり、生存組織を培養することにより、発芽実生や成長した植物体を得ることができる可能性がある。発芽や呈色反応が確認された標本は採集年からの時間経過とともに減少した。発芽もしくは呈色反応が確認され、種子が生存していると判断できたものは合計53種(全種の65%)、107点(全標本の42%)であった。
    以上のことより、標本種子を用いて失われた集団を復元することは種によっては可能であり、標本庫に収められている植物標本は野生植物のシードバンクとして重要な役割を果たせると結論した。なお、現在も発芽試験を継続しており、これらの結果についても報告する予定である。

    シモツケコウホネNuphar submersa Shiga & Kadono(スイレン科)は2006年に新種として発表された栃木県固有の絶滅危惧水生植物である(絶滅危惧ⅠA類)。栃木県中部の河川や水路に生育していたが、圃場整備などによってそのほとんどは絶滅し、現在では3地域(日光市、那須烏山市、真岡市)に群落面積にして合計60㎡残っているに過ぎない。そのような中、生育面積が最も広い日光市では、2009年に移植による系統保存と現状維持地区を設け、保全に配慮した形で圃場整備が実施されることになった。そこで発表者らは有効な保全策の提案を目的に、現存する株の網羅的な遺伝子型決定(ユビキタスジェノタイピング)を行い、各集団の個体数や遺伝的多様性を評価した。
    サンプルは、0.5m間隔で採集し(日光市202株、那須烏山市58株、真岡市7株)、マイクロサテライトマーカー12遺伝子座を用いて遺伝子型を決定した。その結果、3集団とも遺伝子座あたりの対立遺伝子数1.6~1.9、ヘテロ接合度0.11~0.42と低い遺伝的多様性を示したものの、日光市27個体、那須烏山市15個体、真岡市3個体の合計45個体を識別することができ、遺伝子型多様性は集団内に維持されていることが明らかになった。また、各集団はそれぞれ同程度、遺伝的に分化しており(Da=0.38~0.44)、集団サイズが小さく個体数が少ない真岡市の集団も系統的に重要であることが明らかになった。
    日光市での圃場整備を遺伝的多様性の面から評価すると、現状維持地区を設けたことにより20個体を残すことができた反面、残念ながら移植によって複数個体が失われていた。このようにユビキタスジェノタイピングは保全事業のチェック方法としても有効であった。この他にも現地での保全活動について紹介し、ユビキタスジェノタイピングに基づく保全活動の有効性を議論したい。

    シダ植物の博物館標本胞子の生存可能性を評価するために、昨年度標本胞子の発芽試験を行った種に加えて、新たにスギナ(トクサ科)、オオハナワラビ(ハナワラビ科)の2種を研究対象種に加えた。2021年度は、大阪市立自然史博物館と新潟大学の植物標本庫に収蔵されている標本よりそれぞれ胞子(以下、標本胞子と呼ぶ)を新たに得た。
    また、胞子の生理活性を評価するために、オオハナワラビ20標本、スギナ18標本、ゼンマイ19標本に標本胞子に対して、MTTを用いたテトラゾリウム染色試験を行った。オオハナワラビは、標本作成からの経過時間が183ヶ月(15年)の胞子の生存率が85.5±1.7%と最も高く、446ヶ月(38年)経過した胞子の生存率が37.9±4.2%と2番目に高かった。それ以外の年代の胞子は生存率が10%を下回っており、6標本においては0%であった。スギナやゼンマイでは、標本作成からの経過時間が30年から40年にかけて生存率は緩やかに減少した。しかし、各種の幾つかの古い標本では過剰に染色される胞子も確認され、胞子の生存を過剰評価している可能性が考えられた。
    車軸藻類については、昨年度に続き野外採集を行い発芽試験のための標本を増強するとともに、昨年度と今年度作製したシャジクモの標本(群馬県水田産1集団、新潟県ため池産2集団)より卵胞子を採取し、発芽試験(標本卵胞子およびコントロールデータのための非標本卵胞子を各50粒播種、6反復実施)を行った。その結果、新潟県産2集団の標本卵胞子において発芽が認められた(0.3±0.82%~2.7±0.01%)。これまでの研究で卵胞子に乾燥耐性があることは一部の種で示されていたが、標本の卵胞子に着目して発芽を確認したのは本研究が初めてである。今後、車軸藻類においても博物館標本の利用可能性を検討する価値があることを示すデータが得られた。

    On mycology, research activities of citizen scientists have great importance. We tried to enhance the function of natural history museums as support basis of their activities. 1. We made database of Dr. Hongo's mycological collections, and digitized their images, for utilize them for educational tools and materials for modern studies of both taxonomy and ecology. We have got some important results by our research partners in Boletaceae and Amanitaceae. 2. We conducted many scientific seminars for citizen scientists, and we supported series of mycological exhibitions of local museums. 3. We have got good results in mycological DNA barcoding from modern samples and, partly from old Hongo's collection. From these activities, we summarized and reviewed the relation ship between citizen scientists and museums, with special reference to importance of curatorship and open database .

    I investigated the enzyme activity of the embryo and germination test of herbarium specimens's seed of endangered plant of 555 specimens of 117 species. As a result, the seeds of 235 specimens (42%) of 73 species (62%) were alive. The oldest survival specimen seed was Saussurea pulchella (Fisch. ex Hornem.) Fisch. that has been collected 87 years ago.
    I alos examined the effect of specimen preparation method has on the viability of the seed. Specimen seeds subjected to drying treatment at a temperature lower than 40 degrres C showed a high survival rate.
    And I compared the genetic diversity of seedling populations of specimens of Vincetoxicum pycnostelma Kitag. and the wild populations from which the specimens were collected. Genetic diversity of the seedlings were significantly lower than that of wild populations, but the alleles which were lost from the wild population were confirmed.

    I investigated morphological, ecological, and genetical features of Nuphar japonica DC., N. oguraensis Miki, and N. submersa Shiga & Kadono to assess evolutionary process of leaf morphology, life form, and heterophylly in the genus Nuphar Sm.
