    A simple, rapid and accurate evaluation of the sugar uptake rate of suspended plant cells from culture media was developed with the predicted sugar contents measured by mid-infrared spectroscopy using a Fourier transform infrared (FT-IR) spectrometer equipped with an attenuated total reflectance (ATR) accessory. We performed plant cell cultivation with Nicotiana tabacum cv. Bright Yellow No.2 (TBY-2) in culture media, which had various combinations of glucose, fructose and sucrose concentrations at the initial stage, and measured simultaneously each sugar content in the medium by the FT-IR/ATR method. By applying a logistic function to the predicted sugar contents and cell density in the medium during cultivation, the specific sugar uptake rates by the suspended TBY-2 cells were easily and continuously obtained. Thus the kinetic sugar uptake phenomena by the TBY-2 cells were well confirmed overall using the developed method. Additionally it was found that the fraction of sucrose of the initial total sugar content might kinetically affect the sugar uptake process and cell growth. Also, the relationship between the nondimensional cell density and sucrose content could be classified into three groups on the basis of the initial fraction of sucrose.

    In this work the use of Fourier transform infrared (FT-IR) spectroscopy coupled with an attenuated total reflectance (ATR) accessory was applied to determine the polymerization degree of straight-chain oligosaccharides. Glucose (G1) was used as monosaccharide and maitooligosaccharides (G2 similar to G7) that from two to seven glucose polymerized were used as straight-chain oligosaccharides. Seven kinds of same concentration sugar solution (G1 similar to G7) were prepared and their infrared spectra were measured. It was, shown that the shape of spectra in fingerprint region for G5, G6 and G7 were similar. The partial least-squares (PLS) regression was used to determine polymerization degree of maltooligosaccharides from spectra in fingerprint region. The number of C-O-C group was used instead of polymerization degree because C-O-C group forms additionally on polymerization of sugars. Regression analysis against the calculated value gal e a correlation coefficient of 0.999. In order to apply this spectroscopy to on-line prediction of individual sugar concentrations in mixtures, curve-fit of the fingerprint region was attempted to extract the absorption bands affected by polymerization, and three component bands were extracted. The multiple linear regression was used to determine polymerization degree of maltooligosaccharides from spectra in fingerprint region. The number of COC group was predicted by the absorbance of spectra of maltooligosaccharides at three component bands. The comparison between predicted values of the number of C-OC group and the values calculated by preparation of materials showed good agreement. Especially, accurate agree ment was shown for maitooligosaccharides from G1 to G.

    To obtain the respiratory characteristic of agricultural product, the controlled atmospheric storage was conducted by using the desiccator at 5°C, 10°C and 25°C. As the experimental sample, mature green tomatoes and red ripe tomatoes were used. In a closed system, ripening was inhibited for tomato stored in low oxygen atmosphere, and the pseudo-periodic stop was observed at the critical respiration. It was confirmed that at 5°C tomatoes suffered chilling injury in the low oxygen atmospheric storage. Moreover, the oxygen permeability of film package was designed from the critical respiration rate calculated from the data by using the desiccator and it was examined on the effective storage desiccator. The results indicated that the change in surface color of tomato in the packaging film was inhibited. And it was confirmed that both the desiccator and film in the same storage condition showed the same tendency in the storage effect. Therefore, it is also confirmed that the packaging film with oxygen permeability 4, 300ml/m<sup>2</sup>·24h could be applied for the agricultural products.

    An experimental storage system for agricultural products to control temperature and gas composition was constructed. The experiment in which tomato storage characteristics at 25°C under normal atmospheric condition could be obtained with high precision was made by using this apparatus. The immature tomatoes, which typically showed the post-ripening behavior, were used as the experimental samples. For the storage characteristics, O<sub>2</sub> and CO<sub>2</sub> concentrations, total pressure, temperature, and relative humidity were continuously measured while the ethylene and alcohol concentration were determined by inner gas sampling. Surface color and the sugar component changes were obtained as the individual characteristics of tomato. As a result, the O<sub>2</sub> and CO<sub>2</sub> respiration rates reduced largely, and the ethylene evolution highly increased with post-ripening. The surface color also dynamically changed. Moreover, glucose, fructose, sucrose contents in tomato measured by FT-IR/ATR (Attenuated Total Reflection) method before and after storage did not change with post-ripening.

    Evaluation system of fruit appearance, available on Internet, was constructed by using a Java applet to analyze the shape and color that are the main factor of a fruit quality. Since quantified appearance data of fruits, that had been very difficult to calculate heretofore, were easily created and accumulated to the database by this system, the database of fruit appearance was automatically constructed.<br>In this study, the fruits of grape 'Aki Queen' was selected as a sample and its shape and color was analyzed by this system. As a result, the determination of standard shape was made and the temporal change of coloring was also clarified through the quantification process of the shape and color. From these, this system was shown to be useful to the appearance evaluation of a fruit, where not only the binding of the evaluation standard is difficult but also uniform evaluation is difficult to make.

    Considering the surface color change in ripening of tomatoes the color appearance by the production of pigments, the purpose of this study is to examine the color appearance mechanism by measuring the surface color change, to find the relationship between the color development and the production of pigments. As the results, it was found the surface color changed from the special points upon the radial wall of the pericarp. Next, the changing rate of the chlorophyll and the lycopene were calculated by assuming that the carotene content was constant. The calculated values showed the similar tendency to the extracted values of the pigments. This result is suggested that the hue was suitable for the analysis of the production of pigments.

    Color image processing system was constructed to manage the surface color of agricultural products, and several problems in measuring the surface color was examined. Two problems, that is, the setting of condition of illumination which is also an issue in general image processing, and the difference of the color sensation between the visual sense of human and the sense of camera, were dealt with HSL color space. On the problem of illumination, the influence of the quantity of light and the camera angle were examined, and it was confirmed that hue (H) and saturation (S) were not affected by uneven illumination. By the experiments using ripe bananas and immature tomatoes as samples, it was found that the shade of the surface color during post-ripening appeared on the H, and the shade of the surface color in later ripeness appeared on the S.
